Abstract

The aim of the research was to determine the morphology of Simplicillium sp. which can be used as a teaching media in the identification of pathogenic fungi, useful in practical technology for the propagation of biological agents. The research was carried out in the microbiology laboratory at Universitas KH. A. Wahab Hasbullah Jombang, June – August 2023. This type of research is qualitative which aims to determine the morphological characteristics of the fungus Simplicillium lanosoniveum CG888 from the collection of the Faculty of Agriculture UNWAHA as a data source. Data collection methods include observations carried out macroscopically and microscopically, and documentation. Data obtained from observations are compared with existing literature in books and supporting literature to identify the data. The results showed that the colonies of the fungus S. lanosoniveum were white on the upper surface, and yellowish on the bottom. The growth of the colony forms a circle, the edges are white with the growth of new hyphae. The texture of the colony is like wavy cotton with thick mycelia. This fungus has hyaline and septate hyphae. They are branched and tend to be arranged regularly, tapering towards the tip, and have phialids. Phialids produce conidia attached to their tips. Conidia are transparent, small and round in shape. Simplicillium lanosoniveum CG888 has chitinase enzyme activity in it. It is hoped that this teaching media can help students to increase their knowledge of the identification and propagation of fungi which act as biological agents.

ABSTRAK

Tujuan penelitian untuk mengetahui morfologi Simplicillium sp. yang dapat dijadikan sebagai media ajar dalam identifikasi cendawan patogenik berguna pada praktikum Teknologi perbanyakan agens hayati. Penelitian dilaksanakan di laboratorium mikrobiologi Universitas KH. A. Wahab Hasbullah Jombang, bulan Juni – Agustus 2023. Jenis penelitian bersifat kualitatif yang bertujuan untuk mengetahui karakteristik morfologi cendawan Simplicillium lanosoniveum koleksi Fakultas Pertanian Unwaha sebagai sumber data. Metode pengumpulan data meliputi observasi yang dilakukan secara makroskopis dan mikroskopis, dan dokumentasi. Data yang diperoleh dari hasil pengamatan dibandingkan dengan literatur yang ada pada buku dan penunjang pustaka untuk mengidentifikasi data tersebut. Hasil penelitian diperoleh koloni cendawan S. lanosoniveum pada permukaan atas berwarna putih, dan di bawah kekuning – kuningan. Pertumbuhan koloni membentuk bulatan, bagian pinggir berwarna putih dengan adanya pertumbuhan hifa baru. Tekstur koloni seperti kapas bergelombang dengan miselia yang tebal. Cendawan tersebut mempunyai hifa hyalin dan bersepta. Hifa bercabang dengan letak cenderung teratur, dengan meruncing ke ujung, dan terdapat phialid. Phialid menghasilkan konidium yang menempel diujungnya. Konidia transparan, berukuran kecil dan berbentuk bulat. Simplicillium lanosoniveum CG888 terdapat aktivitas enzim kitinase di dalamnya. Diharapkan media ajar tersebut dapat membantu mahasiswa untuk meningkatkan pengetahuannya terhadap identifikasi dan perbanyakan cendawan yang berperan sebagai agens hayati.
